运动对抑郁大鼠学习记忆能力及海马VEGF表达的影响

摘要


目的:对CUMS(慢性不可预见温和应激)致抑郁大鼠进行有氧运动干预,探讨其对海马血管内皮生长因子(VEGF)表达含量及学习记忆能力的影响研究。方法:选用3月龄成年SD大鼠45只,适应性喂养1周后随机分为:对照组/C组、抑郁模型组/M组、抑郁运动组/E组。C组进行为期8周的自然喂养;M组和E组均进行4周CUMS刺激;E组大鼠在CUMS后进行4周有氧游泳运动干预(60分/天,6天/周)。各组大鼠进行糖水消耗实验、全脑5-HT含量检测验证造模结果。Morris水迷宫实验检测大鼠学习记忆能力。Real-time PCR、Western Blotting检测大鼠海马VEGF表达。结果:(1)CUMS会导致大鼠糖水消耗量的降低以及全脑5-HT含量的下降;(2)Morris水迷宫定位航行实验中,各组大鼠逃避潜伏期逐渐下降。第1-2天各组用时无差异,但自第3天开始,M组逃避潜伏期时长明显长于C和E组(P<0.01);空间探索实验M组在60s内穿越原平台的次数明显低于C和E组(P<0.01)。(3)RT-PCR和WB结果表明,与C组相比,M组大鼠海马VEGF mRNA及蛋白表达均显著性降低(P<0.01);E与M组相比,海马VEGF mRNA及蛋白表达有显著性升高(P<0.01),且与C组无差异(P>0.05)。结论:有氧运动可有效逆转CUMS产生的抑郁症状,并改善抑郁大鼠的学习记忆能力。同时,有氧运动可以上调抑郁大鼠海马VEGF的表达,由此表明海马内VEGF表达含量上调可能是介导有氧运动对抑郁及其学习记忆能力改善作用的神经生物学机制之一。
